About GNE Myopathy

What is GNE Myopathy?

GNE Myopathy is a rare, inherited neuromuscular disorder that affects the muscles used for movement (skeletal muscles). It is caused by mutations in the GNE gene, which is responsible for producing an enzyme called UDP-N-acetylglucosamine 2-epimerase/N-acetylmannosamine kinase. This enzyme is essential for the production of sialic acid, which is a sugar molecule found in the membranes of cells. People with GNE Myopathy experience progressive muscle weakness and wasting, as well as difficulty walking, climbing stairs, and standing up from a seated position.

What are the symptoms of GNE Myopathy?

The most common symptoms of GNE Myopathy include:

-Muscle Weakness and wasting, especially in the lower legs and feet
-Difficulty walking, climbing stairs, and standing up from a seated position
-Frequent falls
-Muscle cramps and spasms
-Joint Pain and stiffness
-Difficulty with fine motor skills, such as buttoning a shirt or writing
-Difficulty swallowing
-Fatigue
-Difficulty breathing

What are the causes of GNE Myopathy?

GNE Myopathy is caused by mutations in the GNE gene, which is responsible for producing an enzyme called UDP-N-acetylglucosamine 2-epimerase/N-acetylmannosamine kinase. This enzyme is essential for the production of sialic acid, which is a sugar molecule found in the cell membranes of muscles. Mutations in the GNE gene lead to a deficiency of this enzyme, which in turn causes the muscles to become weak and eventually leads to GNE Myopathy.

What are the treatments for GNE Myopathy?

Treatment for GNE Myopathy is focused on managing symptoms and preventing further progression of the disease. Treatment options may include physical therapy, occupational therapy, orthopedic devices, medications, and dietary modifications. Physical therapy can help improve muscle strength and flexibility, while occupational therapy can help with activities of daily living. Orthopedic devices, such as braces, can help support weakened muscles and joints. Medications, such as immunosuppressants, can help reduce inflammation and slow the progression of the disease. Dietary modifications, such as a low-sugar diet, can help reduce the risk of complications.

What are the risk factors for GNE Myopathy?

1. Genetic mutation in the GNE gene.
2. Family history of GNE Myopathy.
3. Age: GNE Myopathy is more common in adults over the age of 40.
4. Gender: GNE Myopathy is more common in males than females.
5. Ethnicity: GNE Myopathy is more common in individuals of Ashkenazi Jewish descent.
6. Certain environmental factors, such as exposure to certain toxins or medications, may increase the risk of developing GNE Myopathy.

Is there a cure/medications for GNE Myopathy?

Yes, there are medications and treatments available for GNE Myopathy. Treatment options include physical therapy, medications to reduce muscle spasms, and medications to improve muscle strength. In some cases, surgery may be recommended to help improve mobility. Additionally, lifestyle modifications such as avoiding activities that cause pain or fatigue, maintaining a healthy diet, and avoiding smoking can help improve symptoms.
